Abstract
The present study was aimed at the formulation and evaluation of Spironolactone immediate-release tablets using different polymers in varying ratios to enhance the drug release profile and ensure satisfactory tablet characteristics. Spironolactone is a potassium-sparing diuretic widely used in the treatment of hypertension, edema, heart failure, and other cardiovascular disorders. Immediate-release tablets were prepared by direct compression employing different concentrations of selected polymers to investigate their effect on tablet performance and drug release behaviour.
The prepared powder blends were evaluated for pre-compression parameters such as bulk density, tapped density, angle of repose, Carr's index, and Hausner's ratio. The compressed tablets were further evaluated for post-compression parameters including weight variation, thickness, hardness, friability, drug content uniformity, disintegration time, and in vitro dissolution studies. The results demonstrated that all formulations exhibited satisfactory flow properties and compressibility, while the finished tablets complied with the Indian Pharmacopoeia (IP) specifications for all evaluated quality control parameters.
Among the developed formulations (F1–F9), formulation F3 was identified as the optimized formulation based on its superior drug release characteristics and acceptable tablet properties. In vitro dissolution studies revealed that formulation F3 achieved a maximum drug release of 99.21% within 30 minutes, which was significantly higher than that of the other formulations. The enhanced performance of F3 may be attributed to the optimum polymer concentration that facilitated rapid tablet disintegration and drug dissolution.
In conclusion, the study successfully developed Spironolactone immediate-release tablets with desirable pharmaceutical properties. Formulation F3 was found to be the most promising formulation, demonstrating excellent physicochemical characteristics and rapid drug release, making it suitable for effective immediate-release oral drug delivery.
